Genetic similarity of citrus fresh fruit market cultivars Rev. Bras. Frutic.
Oliveira,Roberto Pedroso de; Radmann,Elizete Beatriz.
The objective of this work was to assess the genetic similarity of the following citrus fresh fruit market seedless cultivars: Lane Late, Navelate, Navelina and Salustiana sweet oranges (Citrus sinensis (L.) Osbeck), Clemenules and Marisol mandarins (C. reticulata Blanco) and Okitsu satsuma mandarin (C. unshiu Marcovitch), and the hybrids Nova [C. clementina x (C. paradisi x C. tangerina)] and Ortanique (tangor probably derived from C. sinensis (L.) Osbeck x C. reticulata Blanco), utilizing isoenzymatic markers. Electrophoresis analysis of proteins extracted from leaf tissues was utilized to detect polymorphisms at ten isoenzymatic systems. Out of 30 alleles, 16 were polymorphic. The Jaccard coefficient was utilized to estimate the genetic similarity...

Iodophanus carneus and I. testaceus (Ascomycota-Pezizales): Independent taxonomic identity or synonymy? A study of their morphology and isozymes Boletín de la Sociedad
Cinto,Isabel E.; Dokmetzian,Diana A.; Ranalli,María E..
The aim of this study was to delimit two Iodophanus species: I. carneus and I. testaceus , based on morphological characteristics and electrophoretic patterns of their intracellular isozymes. Twenty monosporic strains were used, including five belonging to I. granulipolaris as a control. Fourteen isozyme systems were tested, and the five having the best resolution selected: aspartate aminotransferase, esterases, alkaline phosphatase, glutamate dehydrogenase, and superoxide dismutase. These analyses confirmed the similarity between I. carneus and I. testaceus , since they both produced the same band patterns, which were in turn different from the band pattern of I. granulipolaris. So, as we couldn´t find any character wich permit us to classify the isolated...

Is there genetic variation in seedless Argentinean grapefruit? Implications for crop production and conservation JBAG
Chacoff,N P; Souto,C P; Aizen,M.A.; Premoli,A.
Increasing crop genetic diversity has the potential to enhance pollination services, by contributing to the long-term stability of agroecosystems. Different species of the genus Citrus, including grapefruit, are cultivated worldwide. In Argentina, 30% of the total national yield is concentrated in the northwestern part of the country where new cultivars have arisen. We analyzed genetic variation and diversity within and among four plantations of Citrus paradisi for 11 isozyme loci. Although representing single cultivars clonally propagated, within plantations genetic variation was high. Observed heterozygosity ranged from 33 to 36.5%. The number of alleles per locus ranged from 2.4 to 2.6, and polymorphism from 72.7 to 81.8% and for the species in the area...

Isoenzymatic variability in tropical maize populations under reciprocal recurrent selection Scientia Agricola
Pinto,Luciana Rossini; Vieira,Maria Lúcia Carneiro; Souza Jr.,Cláudio Lopes de; Silva,Rainério Meireles da.
Maize (Zea mays L.) is one of the crops in which the genetic variability has been extensively studied at isoenzymatic loci. The genetic variability of the maize populations BR-105 and BR-106, and the synthetics IG-3 and IG-4, obtained after one cycle of a high-intensity reciprocal recurrent selection (RRS), was investigated at seven isoenzymatic loci. A total of twenty alleles were identified, and most of the private alleles were found in the BR-106 population. One cycle of reciprocal recurrent selection (RRS) caused reductions of 12% in the number of alleles in both populations. Changes in allele frequencies were also observed between populations and synthetics, mainly for the Est 2 locus. Populations presented similar values for the number of alleles per...

Isolation and partial purification of beta-galactosidases from cotyledons of two cowpea cultivars Rev. Bras. Fisiol. Veg.
ENÉAS-FILHO,JOAQUIM; BARBOSA,GISLAINY KARLA DA COSTA; SUDÉRIO,FABRÍCIO BONFIM; PRISCO,JOSÉ TARQUÍNIO; GOMES-FILHO,ENÉAS.
Three isoforms of beta-galactosidases were isolated and partially purified from the cotyledons of quiescent seeds of Vita 3 and Vita 5 cowpea [Vigna unguiculata (L.) Walp.] cultivars differing in water and salt stress tolerance. The purification procedure consisted of ammonium sulfate fractionation, acid precipitation, ion exchange chromatography through DEAE-sephadex and affinity chromatography through Lactosyl-sepharose columns. The three isoforms isolated from the two cultivars showed the same chromatographic patterns, same optimum of temperature for enzyme activity assay (60ºC), identical thermal stability up to 50°C, and similar pH optima (3-4). However, they differed from each other in sensitivity towards metal ions and certain chemical agents...

Isozyme variation in four species of the Simulium perflavum species group (Diptera: Simuliidae) from the Brazilian Amazon Genet. Mol. Biol.
Scarpassa,Vera Margarete; Hamada,Neusa.
Electrophoretic studies of isozymes were done with four closely related species of the Simulium perflavum species group (Diptera: Simuliidade) in the Brazilian Amazon, using last-instar larvae collected in the field. Ten enzymes were studied, which yielded 11 loci. Diagnostic loci were not found between Simulium maroniense cytotype D and Simulium rorotaense. Simulium maroniense and S. rorotaense differed from Simulium trombetense by two diagnostic loci (Me and Xdh), and Simulium perflavum differed from the other three species by four diagnostic loci (Me, Xdh, Mdh, and Got). The mean number of alleles per locus ranged from 1.30 to 2.30, the percentage of polymorphic loci ranged from 18.2 to 63.6% and the mean heterozygosity values observed ranged from 0.062...

Mating system in a neotropical tree species, Senna multijuga (Fabaceae) Genet. Mol. Biol.
Ribeiro,Renata Acácio; Lovato,Maria Bernadete.
Senna multijuga is a pioneer tropical tree species that occurs mainly in the Brazilian Atlantic forest. We investigated the mating system of two populations of S. multijuga, one located in a reserve area (RD1) and the other (RD2) about 15 km away. The mating system parameters were estimated using the mixed mating model (software MLTR). The two populations had significantly different outcrossing rates, with population RD2 having a high rate (t m = 0.838) and population RD1, a lower rate (t m = 0.540). The values of t s were different between the two populations and also lower than those of t m. Significant t m - t s estimates indicated that biparental inbreeding contributed to the apparent selfing rate in these populations. The correlation of paternity was...

Phytophthora infestans (Mont.) de Bary. I. Host-Pathogen specificity and resistance components Agrociencia
Lozoya-Saldaña,Héctor; Guzmán-Galindo,Leonel; Fernández-Pavia,Sylvia; Grünwald,Niklaus J.; McElhinny,Elaine.
Abstract The molecular identity of Phytophthora infestans strains infecting potatoes in the Toluca Valley has been documented. Nevertheless, the relationship of the P. infestans genotypes to the host’s genetic resistance is unclear. In order to identify potential host-pathogen specificity, isolates of the oomycete obtained from five potato varieties with differing levels of disease resistance were compared regarding mating type (MT), isozyme genotype for glucose-6-phosphate isomerase (Gpi) and peptidase (Pep), and sensitivity to metalaxyl. Also, host resistance components and specificity on the host-pathogen interaction were assessed twice in detached leaves in the laboratory and once in attached leaves in the greenhouse for three of the varieties. No...

Phytophthora taxa associated with cultivated Agathosma, with emphasis on the P. citricola complex and P. capensis sp. nov. Naturalis
Bezuidenhout, C.M.; Denman, S.; Kirk, S.A.; Botha, W.J.; Mostert, L.; McLeod, A..
Agathosma species, which are indigenous to South Africa, are also cultivated for commercial use. Recently growers experienced severe plant loss, and symptoms shown by affected plants suggested that a soilborne disease could be the cause of death. A number of Phytophthora taxa were isolated from diseased plants, and this paper reports their identity, mating type, and pathogenicity to young Agathosma plants. Using morphological and sequence data seven Phytophthora taxa were identified: the A1 mating type of P. cinnamomi var. cinnamomi, P. cinnamomi var. parvispora and P. cryptogea, the A2 mating type of P. drechsleri and P. nicotianae, and two homothallic taxa from the P. citricola complex. The identity of isolates in the P. citricola complex was resolved...

Relationships among six herbal species (Curcuma) assessed by four isozymes Phyton
Deng,JB; Ding,CB; Zhang,L; Zhou,YH; Yang,RW.
Four isozymes, superoxide dismutase (SOD), polyphenol oxidase (PPO), malate dehydrogenase (MDH) and cytochrome oxidase (COD) were studied for identification of six herbal species (Curcuma L.). All the 37 study specimens produced a total of 168 polymorphism isozyme bands. The genetic distance coefficients (GS) varied from 0.08 to 0.54. The dendrogram, obtained according to the polymorphism isozyme bands by the UPGMA method with the software NTSYS-pc2.1, contributed to improve the resolution of phylogeny. From the dendrogram, it was possible to differentiate between the wild and cultivated specimens of C. longa, and within C. sichuanensis species.
